Effect of Lithium Ion on Cab Gene Expression in Soybean

@EN The expression of cab genes in soybean is regulated by light. The molecular details on how light signal is transduced to activate light-light-induced genes in plant is largely unknown. To see whether phosphoinositide turnover is involved in
the
light signal transduction, the effect of li+, a phosphoinositide metabolism blocker, on cab gene expression was examined. The effect of the other monovalent cations, Na+ and K+, was also examined. When soybean seedlings were treated with Li+,
Na+,
or
K+, the expression of cab genes was substantially decreased, even in the presence of light. Among the monovalent cations, Li+ was the most potent inhiitor of cab gene expression. Identical treatment did not significantly inhibit the expression of
a
housekeeping gene. It was also found that the cab gene expression is subject to osmotic stress. However, the effect of Li+ is more pronounced than the effect of mannitol, implying that the Li+ ion itself has an additional effect in addition to
the
osmotic effect. The strong inhibitory effect of Li+ on cab gene expression might reflect the fact that the phosphoinositide turnover was blocked by Li+ and that phosphoinositide metabolism plays an important role in light signal transduction to
light-regulated genes.
